In vitro models:
In vitro testing methods include cells lines specific for HIV-1 studies using recombinant proviral clones and other models in transformed and primary lymphocytes. Determination of IC-50 for anti-HIV drugs.
Parameters:
- Inhibition of cell death by HIV-1 in MT-2 cells (MTT assays) (co-operation)
- Determination of p24 antigen in culture supernatants of primary T-cells infected with HIV-1 (co-operation)
- Determination of the transcriptional activity of the LTR-HIV promoter by Tat and by TNFa.
- Determinations of anti-HIV compounds targetting Tat/TAR interaction and RNA elongation. CDK9 phosphorylation assays
Anti-HIV-1 activity using VSV-pseudotyped recombinant and HIV recombinant clones
- Reverse transcriptase assays and integration assays
